Plant Cell Vacuoles

چکیده

The vacuole is the largest compartment of a mature plant cell and may occupy up to 95% of the total cell volume. In such mature cells, the cytosol is visible only as a thin layer, which is separated from the cell wall by the plasma membrane, and from the vacuolar sap (cell sap) by the vacuolar membrane (tonoplast). The constituents of the cell sap are mainly inorganic salts and water. The vacuole, therefore, enables the plant to reach a large size and a large surface area by accumulating salts from the environment, which drive further water uptake osmotically. Thus only a minimumof energy is diverted from the energy-consuming synthesis of metabolites. The requirement for cytosolic ions to be in homeostasis is also met by the vacuole as it serves as an internal reservoir of metabolites and nutrients (Matile, 1978; Boller and Wiemken, 1986; Martinoia, 1992). It can also be used for storage of other compounds that would be problematic in the cytosol, such as ions, pigments, storage compounds or toxic substances.

A carboxyl-terminal propeptide is necessary for proper sorting of barley lectin to vacuoles of tobacco.

Barley lectin is synthesized as a preproprotein with a glycosylated carboxyl-terminal propeptide (CTPP) that is removed before or concomitant with deposition of the mature protein in vacuoles. Expression of a cDNA clone encoding barley lectin in transformed tobacco plants results in the correct processing, maturation, and accumulation of active barley lectin in vacuoles [Wilkins, T.A., Bednarek...

Immunological Identification of Proteinase Inhibitors I and II in Isolated Tomato Leaf Vacuoles.

Proteinase inhibitor I has been identified and quantified in isolated vacuoles from tomato (Lycopersicon esculentum) leaves induced to accumulate inhibitors either by wounding or by supplying excised leaves with the wound hormone, proteinase inhibitor-inducing factor. Proteinase inhibitor II was also identified in the vacuoles but not quantified. Control vacuoles were prepared from unwounded pl...

Protein dynamics and proteolysis in plant vacuoles.

Plant cells cannot live without their vacuoles. The tissues and organs of a plant contain a wide variety of differentiated and specialized vacuoles -- even a single plant cell can possess two or more types of vacuoles. Vacuolar proteins are encoded by nuclear genes and synthesized in the cytoplasm. Their transport into the vacuolar compartment is under cytoplasmic control. Transcription seems t...

Plant vacuole morphology and vacuolar trafficking

Plant vacuoles are essential organelles for plant growth and development, and have multiple functions. Vacuoles are highly dynamic and pleiomorphic, and their size varies depending on the cell type and growth conditions. Vacuoles compartmentalize different cellular components such as proteins, sugars, ions and other secondary metabolites and play critical roles in plants response to different b...

Exoamylase activity in vacuoles isolated from pea and wheat leaf protoplasts.

Vacuoles isolated from pea (Pisum sativum), and wheat (Triticum aestivum) leaf protoplasts contained considerable activities of electrophoretically highly mobile exoamylases. Vacuoles from spinach (Spinacia oleracea) leaf and photoautotrophic Chenopodium rubrum suspension culture cell protoplasts were devoid of amylolytic activity. Endoamylase activity was in all cases associated primarily with...

The relationship between vacuolation and initiation of PCD in rice (Oryza sativa) aleurone cells

Vacuole fusion is a necessary process for the establishment of a large central vacuole, which is the central location of various hydrolytic enzymes and other factors involved in death at the beginning of plant programmed cell death (PCD). In our report, the fusion of vacuoles has been presented in two ways: i) small vacuoles coalesce to form larger vacuoles through membrane fusion, and ii) larg...
